I'd start with using DateDiff for example:
DateDiff('day', [Enquiry], [Responded])
DateDiff('day', IFNULL([Responded], [Enquiry]), [Assigned])
You can then adjust the 'day' element to suit your needs, for example set to 'minute' instead

But its giving me wrong output when there is difference between 2 or 3 days.
Lets say my StartDate is 11/12/2017 15:56:19 and EndDate is 11/26/2017 11:21:58
Default property on Number Format is set to dd Hh:Nn:Ss
Output is showing as 12 19:25:39
Let me know if there is any mistake in calculations or Number Format.
