Calculated fields are just like any another column in the database - and it is computed run time - but you can specify what goes into this column. When the report is run, it calculates this field field for each row.
Sorry, the last part of your question is severely flawed and lacks DB understanding I think. Please elaborate, what you are trying to accomplish.
These values are generated by tableau when the report is run, I need to store the values in a calculated field for calculation purpose. Is there any direct way I can do that? For example if I want amount in asia/total
GO To WORKSHEET>Export>Data.. to copy all whatever is in view and then you can use it as a new connection and use data.
Worksheet>Copy>Copy>Crosstab and paste as new data source

I am looking to use the Measured Values in Calculated field so that the report can be run without the need to connect to a new data source every time.
A calculated field is like an equation in Excel - it will give you the result based on the available data, but it won't "store" the result for later.
To use these values in a calculated field, simply reference them - so if you're displaying Sales data then you can SUM([Sales]), or MAX[(Sales)] or any other sort of aggregate calculation, or you can simply say [Sales] if you want to access the value in each row ... it just depends on what you want to achieve as an end result.
