Download the datasource from your server as a local copy, edit the datasource details and republish it using the same name as the existing published connection.

If i am understanding your question correctly.
You have, say 10 data sources which you published it to your Tableau server. as DS1, DS2, DS3....DS10. with Database connection as ABC1.DB
Now, Your organisation decided to change the Database from ABC1.DB to ABC2.DB
If yes, I don't see any issue here. You do have edit Datasource connection option for the published data sources. We are using Tableau server 10.0.1 and here is the snapshot.
You can edit the connection details and point to the new server name.
