Yes you can...the great thing about Tableau is that although it builds (reasonably) intelligent ToolTips for you...you can also drag any fields you want into the tooltip. In the attached I have shown what Tableau builds by default, and then in the 'Different Formatting per Measure' tab I've dragged the measures in (onto the ToolTip tile) and then done a bit of manually editing of the tooltip (if you click on the ToolTip tile it opens the text box where you can edit the text). I've brought in the 2 measures in the view and a 3rd one not in the Viz at all.
For some really fancy control, you may also like some of the tricks I've used here (on both formatting and colouring +ve/-ve)

Regarding your example what I would like to achieve is:
When you hover over Sales it's showing sales only in the Tooltip
When you hover over Profit it's showing profit only in the Tooltip
Sales and Profit are nice measures to compare, but sometimes we don't want this.
So the final solution will depend on your Viz/how many measures...etc. We do have that behavior in the 'Default ToolTip' tab, and by using the Formatting (in the Pane) tab I can control how each displays (in terms of number formatting)...in the updated workbook I've given Profit 2 decimal places
For total control, one way is to use a Dual Axis (and Sync Axis). If you look at my 'Dual Axis' tab I have done this, and I get a ToolTip per Axis, where I've changed the text to show this.
If this doesn't work for you, can you post a picture of your chart so I can see the number of measures/arrangement...etc. and see if I an come up with something!

Is it possible to add all measure names and measure values in tool tip without adding those individual measures to tool tip shelf?
In the above screenshot, I have added each measure to tool tip shelf. If I drop
<Measure Names>: <Measure Values>, only its relevant values are displayed like you said.
Is it possible to make the relevant measure and value bold in the screenshot approach above?
Say if you hover over profit, the tooltip should show profit and its value as bold and others normal..Similarly if you hover over sales, sales value and name should be bold. Is it possible?
I afraid not.
The only way to highlight in bold the relevant measure is to Bold the text in the tooltip for <Measure Name>: <Measure Value>
and add all the other fields below.

I did consider the same approach you suggested yesterday. Drawback is the the measure will be duplicated in tool tip meaning it will appear twice.
Say if you hover over sales <measure name>: <measure value> will show sales in addition to the individual field we dragged and dropped - Sales: <SUM(Sales).
So this users may not want to see.
