Yes, you are correct in assuming that if the DB2 database has been refreshed with data on Monday, when you run the report on Tableau Server next day, or straight after the database has received the new data, it should show up on the reports. This is because Tableau queries the database every time you run the report or make a change, so if the connection is 'Live', you are always looking at the latest data.

One small info needed on "Live Connections".
1) Does it work with MYSQL/Oracle DB. We found it does not work with MYSQL.
2) Every time we open the dashboard it is asking for DB password before opening the dashboard. Is there any way to connect live without asking end user to supply for DB password.
