To restate what you're asking, you want to have a single selection mechanism to select a State to inherently select the Region, but to exclude the chosen State?
Is there a reason you can't just use 2 Quick Filters with the "exclude" functionality provided by Tableau? (One to select Region to show, and one to select State to Exclude)
Your original question is possible, but I'm finding it a little hairy. I thought I'd see if this would suffice first.

I would like to use dynamic filters rather than asking my users to click on all the selections. Is there a way?

There wouldn't be need to click on "all the selections."
They would choose the Region they want to show from the dropdown. (Region quick filter).
They would choose the State they want to exclude from the dropdown/checkbox list (State quick filter).
So in 2 clicks, you select the region and exclude a state in that region.
I've attached the workbook with what I'm describing on Sheet 6.
Thank you for the example. But, I would like to apply the same filter on another cross tab to include that value.
In the example I have procided in the workbook, When I select East,. Maryland, it should be excluded from sheet 6 but sheet 7 should show East and only maryland.
