You are able to publish both TDE and TDSX files to the Tableau Server.
A TDE is a data extract, i.e. a file that just contains the Data.
A TDSX file, contains both the data (i.e. the TDE), the data connection information (i.e. where the data came from), and any additions to the data (i.e. calculated fields, parameters, etc)
Therefore, if you want to be able to refresh the extract, or have things like calc fields in your data source, you should publish a TDSX

I would take a look at the following docs
The general steps are (for publishing extracts to the server)
1. In Tableau Desktop, connect to Data source
2. Tidy up data source (field names, unwanted fields, calculated fields, etc)
3. Create Extract
4. Publish to Server, and assign refresh schedule.
Things to think about, are publishing to a project which only gives permissions to the correct groups / users to access the data source.
Also, if you are new to Tableau Server, you may want to take a look at enrolling on the Server Admin course, as these sorts of topics are covered on it.
