Are you trying to calculate a value for 'Male' and 'Female' or do you need to manipulate your source data to match it?
Should your 'Under 35' column be the sum of Female + Male under 35?
Can you please attach some sample data in a Tableau Packaged Workbook?

Actually i am looking for 2 columns. One is Gender ie. Male , Female(2 columns Only) and another as Age with 3 column like shared.
All 5 columns will be independent.
Without the requested Tableau Packaged Workbook or the actual definitions of how you want to calculate Male and Female , my advice will be general.
You would need a calculated field for each 'column' then you will need to remove the Q141 and Q140 dimensions from the columns shelf and instead replace it with Measure Names from dimensions and Measure Values from Measures and select the new calculated fields you've created.
I.e. a calculated field for Female;
SUM(IF Q141 = 'Female' then [ValueField] END)
SUM(IF Q141 = 'Male' then [ValueField] END)
SUM(IF Q140= 'Under 35' then [ValueField] END)
Repeat for 35-54 and 55+ as per Under 35 example.
