Hey Priyanka, I'll shoot from the hip here, and ask: Is the path you used to connect to the file the full UNC path? Server will need your connection to say: Connect to "\\fileserver\myshare\myfile.txt" rather than a mapped network drive such as "H:\myshare\myfile.txt".
The other problem might be that "Include external files" was checked in the Publish dialog in Desktop. It shouldn't be--leave it blank if you want Server to refresh the extract from the shared file on a schedule.
If that doesn't immediately resolve this--what is the problem you are seeing? Does Tableau Server report an error when it tries to refresh the extract from the file, or are you unable to get it to allow you to schedule the refresh from Desktop?

Besides what Matt said, check out I Have Wee Data – Microsoft Access and Tableau | Drawing with Numbers. MS Access is also a file-based source so what is written about what you need to do (and *not* do) for MS Access also applies to text & Excel files.
