What your asking for is not possible out of the box. The easiest way to achieve this would be to connect the Tableau report directly to the database and use a live connection.
If that's not an option, you would need to use some of script to monitor for changes in the Excel file then use something like the extract API to create a TDE file automatically.

You can use an ETL tool to accomplish this. Something like Pentaho Data Integration CE 6.0+ along with the Tableau TDE Output plugin. You'd create a job and use the "Wait For File" step, then you'd have a subsequent transformation that loads the Excel file using a Microsoft XLS Input step and output to the TDE Output step.
