You will likely want to reshape your data anyway to look like this

Then if the times are really times in Excel (rather than strings), you should be able to analyze the data in Tableau. The online support has some great training videos
Even after that "Time" column not coming under measure, in that case how can I get the measure form that column.

No, that's no good - I need it from your original Excel file. Your time could be a real time recognized by Excel or a text string. Ideally attach a packaged Tableau workbook.
@Alex: I got this by copying the posted table, pasting it into a new Excel workbook, then copying that data and pasting it into Tableau).
@Yuvaraj: I'm imagining by saying you can't turn Time into a measure you're thinking that you need it in the Measures section. That makes me think you're not familiar with the Continuous/Discrete distinction that is actually the difference between blue pills and green pills. These two posts are really helpful.
Here's what I did in Tableau with the imported data:
- On a new worksheet, right-click & drag Date onto the Columns Shelf, Tableau offers a dialog with aggregation options.
- Choose the second (green) DAY(Date) option.
- Right-click and drag Time onto the Rows shelf, Tableau again offers a dialog.
- Choose the second (green) MINUTE(Time) option. Tableau now displays a scatterplot with the Shape Mark Type.
- On the Marks Card, click on the combo box for the Mark Type and change it to Line.
Then you can right-click on the Time axis to format it as desired, for example to set the Tick mark locations, etc.

My time data is a string, but I have changed the data type to Data/Time format in tableau.
