I'm not sure I understand your aim here. Are you attempting to limit the values displayed as options in a quick filter?
e.g., If you have a dimension with 3 values "High", "Medium" and "Low" but you know your users never need to see "Low" and should in fact not see "Low" as an option, you can "remove" that value from the quick filter...
The solution to that would be to duplicate the dimension field, drag the duplicated dimension to the Filter shelf (in addition to the original) and exclude "Low." Now, you can expose the original filter, but select "Show Relevant Values only" and the users will see only "High" and "Medium" as options.
Is that what you are getting at?
To answer your second question, you can set the filter to be a wildcard match, or a custom value list. wildcard will return all values matching the substring you provide, custom value will return them in a multi-selectable list and you can further exclude from there.
